Media in the United States have reported that the star of Mrs Doubtfire was admitted to hospital on Thursday with heart problems.
Mr Williams, 57, said in a statement on his website that the next four performances of his "Weapons of Self-Destruction" show in south Florida have been postponed.
"Williams was experiencing shortness of breath and is currently being evaluated by doctors who are recommending a week of rest," the statement read.
Mr Williams is "currently at a Miami hospital with heart issues and could be laid up for a while" the Miami Herald newspaper reported, quoting anonymous sources.
He was in the unnamed hospital's intensive care unit, the Los Angeles Times added.
Mr Williams won a best supporting actor Oscar for his role in the 1997 film Good Will Hunting and has been nominated for an Academy Award for three other movies. These include Good Morning Vietnam, The Fisher King, and Dead Poets Society.
He has been on an 80-city comedy tour since September performing to sold-out crowds, his website said.
According to his website, tickets for his New York dates - his first in the city since 2002 - sold out in under 10 minutes.
He will appear in the sequel to Night at the Museum, Battle of the Smithsonian, later this year as President Teddy Roosevelt.
